Biophysicists unravel secrets of genetic switch

ScienceDaily (Aug. 30, 2012) ? When an invading bacterium or virus starts rummaging through the contents of a cell nucleus, using proteins like tiny hands to rearrange the host's DNA strands, it can alter the host's biological course. The invading proteins use specific binding, firmly grabbing onto particular sequences of DNA, to bend, kink and twist the DNA strands. The invaders also use non-specific binding to grasp any part of a DNA strand, but these seemingly random bonds are weak.

Emory University biophysicists have experimentally demonstrated, for the fist time, how the nonspecific binding of a protein known as the lambda repressor, or C1 protein, bends DNA and helps it close a loop that switches off virulence. The researchers also captured the first measurements of that compaction.

Their results, published in Physical Review E, support the idea that nonspecific binding is not so random after all, and plays a critical role in whether a pathogen remains dormant or turns virulent.

"Our findings are the first direct and quantitative determination of non-specific binding and compaction of DNA," says Laura Finzi, an Emory professor of biophysics whose lab led the study. "The data are relevant for the understanding of DNA physiology, and the dynamic characteristics of an on-off switch for the expression of genes."

C1 is the repressor protein of the lambda bacteriophage, a virus that infects the bacterial species E. coli, and a common laboratory model for the study of gene transcription.

The virus infects E. coli by injecting its DNA into the host cell. The viral DNA is then incorporated in the bacterium's chromosome. Shortly afterwards, binding of the C1 protein to specific sequences on the viral DNA induces the formation of a loop. As long as the loop is closed, the virus remains dormant. If the loop opens, however, the machinery of the bacteria gets hi-jacked: The virus switches off the bacteria's genes and switches on its own, turning virulent.

"The loop basically acts as a molecular switch, and is very stable during quiescence, yet it is highly sensitive to the external environment," Finzi says. "If the bacteria is starved or poisoned, for instance, the viral DNA receives a signal that it's time to get off the boat and spread to a new host, and the loop is opened. We wanted to understand how this C1-mediated, loop-based mechanism can be so stable during quiescence, and yet so responsive to switching to virulence when it receives the signal to do so."

Finzi runs one of a handful of physics labs using single-molecule techniques to study the mechanics of gene expression. In 2009, her lab proved the formation of the C1 loop. "We then analyzed the kinetics of loop formation and gained evidence that non-specific binding played a role," Finzi says. "We wanted to build on that work by precisely characterizing that role."

Emory undergraduate student Chandler Fountain led the experimental part of the study. He used magnetic tweezers, which can pull on DNA molecules labeled with miniscule magnetic beads, to stretch DNA in a microscope flow chamber. Gradually, the magnets are moved closer to the DNA, pulling it further, so the length of the DNA extension can be plotted against the applied force.

"You get a curve," Finzi explains. "It's not linear, because DNA is a spring. Then you put the same DNA in the presence of C1 protein and see how the curve changes. Now, you need more force to get to the same extension because the protein holds onto the DNA and bends it."

An analysis of the data suggests that, while the specific binding of the C1 protein forms the loop, the non-specific binding acts like a kind of zipper, facilitating the closure of the loop, and keeping it stable until the signal comes to open it.

"The zipper-like effect of the weaker binding sites also allows the genetic switch to be more responsive to the environment, providing small openings that allow it to breathe, in a sense," Finzi explains. "So the loop is never permanently closed."

The information about how the C1 genetic switch works may provide insights into the workings of other genetic switches.

"Single-molecule techniques have opened a new era in the mechanics of biological processes," Finzi says. "I hope this kind of experiment will lead to better understanding of how our own DNA is compacted into chromosomes, and how it unravels locally to become expressed."

Other authors on the paper include Sachin Goyal, formerly a post-doc in the Finzi lab; Emory cell biologist David Dunlap; and Emory theoretical physicist Fereydoon Family. The research was funded by the National Institutes of Health.

Delaware secret court arbitration unconstitutional

DOVER, Del. (AP) ? A Delaware law allowing Court of Chancery judges to preside over secret arbitration in business disputes is unconstitutional, a federal judge ruled Thursday.

The ruling came in a lawsuit filed by the Delaware Coalition for Open Government, which argued that the secret proceedings violated the rights of citizens to attend judicial proceedings and access court records.

In a 26-page ruling, U.S. District Court Judge Mary McLaughlin agreed that the Chancery Court, which often presides over high-profile business disputes involving some of the world's largest corporations, cannot conduct secret arbitration.

McLaughlin noted that under the arbitration process, a sitting judge hears evidence, finds facts and issues enforceable orders.

"The court concludes that the Delaware proceeding functions essentially as a non-jury trial before a Chancery Court judge," wrote McLaughlin, who heard oral arguments in February. "Because it is a civil trial, there is a qualified right of access and this proceeding must be open to the public."

David Finger, an attorney representing the open-government coalition, said the ruling was a reminder of the importance of transparency in all branches of government.

Gov. Jack Markell and Chancellor Leo Strine Jr. referred requests for comment to Lawrence Hamermesh, a Delaware law professor who assisted in representing the state. Hamermesh said in a prepared statement that state planned to appeal because of the economic importance of providing businesses with "cost-effective ways to resolve disputes."

The law allowed secret arbitration in business disputes involving claims for monetary damages exceeding $1 million. Attorneys for the state argued that the secret arbitration made the court more efficient and generated revenue for Delaware, the corporate home for thousands of U.S. companies.

But court officials disclosed earlier this year that only five secret arbitrations had been held, all lasting only one day and generating only $60,000 in revenue for the court.

Under the law, the court charged $12,000 for filing an arbitration petition and a daily fee of $6,000 for each day after the first day that a judge was engaged.

Because the proceedings were cloaked in secrecy and not even put on the court docket, the public had no way of knowing how often judges were engaged in arbitration. The coalition filed its lawsuit shortly after two companies disclosed in filings with the Securities and Exchange Commission that they were seeking arbitration in a failed merger. Without the SEC filings, the public may have never known about the case.

Several media organizations, including The Associated Press, The New York Times, The Washington Post and the Reporters Committee for Freedom of the Press, submitted a brief supporting the coalition.

The corporate law section of the Delaware State Bar Association, along with the New York Stock Exchange and Nasdaq, submitted briefs supporting the arbitration law.

Many suspected ineligible CO voters US citizens

DENVER (AP) ? Nearly a third of people whose citizenship and right to vote were questioned by Colorado's secretary of state are actually U.S. citizens, election officials said Wednesday, prompting Democrats to question the motives behind the effort to clean up voting rolls as a tightly contested presidential election approaches.

Earlier this month, Republican Secretary of State Scott Gessler sent letters to nearly 4,000 people questioning their citizenship as part of a plan to have them voluntarily withdraw or confirm their eligibility to vote.

State officials were able to run 1,400 of those names through a federal immigration database and found that more than 1,200 were U.S. citizens. So far, they've found none who are non-citizens and registered to vote.

Martha Tierney, an attorney for the Colorado Democratic Party, told election officials during a meeting Wednesday that they were wasting their time on a small group of voters instead of focusing on ensuring a fair and accurate fall election.

"This is a witch hunt and you should be embarrassed that you're going down this road," she said.

Gessler's office plans to release updated figures on Thursday detailing how many of the 4,000 people responded directly to affirm their citizenship or withdraw their voter registration. He said no further action will be taken involving people who did not respond to the letters.

Democrats have criticized the effort to correct the voting rolls and said it could disenfranchise legal voters or make it difficult to exercise their right to vote. More than three-quarters of the letters went to Democrats and independent voters.

Gessler denies any political motivation and insists his goal is to maintain accurate voter rolls. His office said it did not look at party registration when sending the letters.

Gessler spokesman Rich Coolidge said that ensuring only eligible voters cast ballots is an important component of running a successful election.

"As the state's chief election official, he is obligated to make sure that only eligible voters are casting ballots," Coolidge said before the hearing. "We identified a vulnerability in the system, we identified people who exploited, or accidentally exploited that vulnerability, and we're going to shut down that loophole."

Still, critics have questioned Gessler's political motivations in a year where both major parties see Colorado as key to winning the presidential election. Control of the state Legislature and competitive congressional races are also at stake.

Across the country Republicans have aggressively pursued initiatives to verify voters' citizenship, particularly in swing states, much to the ire of Democrats who worry that key parts of their base ? Latinos and seniors ? are likely to be disenfranchised.

Election chiefs in Iowa, Michigan, New Mexico and Ohio ? all expected to be competitive in November ? joined Colorado and other states asking the federal government for access to the database to verify citizenship. Colorado got access to the database last week.

Samantha Meiring, 37, a Colorado voter whose status was questioned, waved a letter as she told election officials Wednesday that she was speaking "as an immigrant and U.S. citizen who got a lovely little letter in the mail."

"I find it absolutely ridiculous that a U.S. citizen is being asked to jump through additional hoops to exercise a right to vote," said the registered Democrat, a South African immigrant who became a U.S. citizen in 2010. "I think you're chasing people that don't need to be chased."

A total of 1,566 letters went to Democrats and 1,794 went to unaffiliated voters. Another 486 letters were sent to Republicans.

Using information from the department of motor vehicles, Gessler identified people who once presented documents showing they were not citizens, such as a green card, when applying for a driver's license. It's unclear whether those people registered to vote by accident while getting a driver's license or when approached by someone as part of registration drive.

Gessler's office provided The Associated Press with the party affiliation of people who received the letters but denied a request to see their names, citing an ongoing investigation.

Democrats and voting advocacy groups said Gessler has shown no proof of widespread fraud and overreached by sending the letters.

"They basically sent out the functional equivalent of an email blast," said Mark Grueskin, an attorney who represents Democrats on election issues.

The American Civil Liberties Union in Colorado said at least 25 eligible voters who got the letters have contacted the organization and the number is growing.

Democrats do not want Gessler to hold hearings for the fewer than 200 remaining people whose citizenship status hasn't been verified on the federal immigration database. They say further hearings are unnecessary because county clerks already have the power to handle challenges to voter registrations.

If Gessler's plan to hold hearings is approved by his office, people could start receiving a new round of letters as soon as next week notifying them that their citizenship is in doubt.

Deputy Secretary of State Suzanne Staiert said the goal is to keep accurate voter rolls, and that officials are working in the best interest of immigrants who may be unaware that they're breaking the law by being registered to vote or by voting.

"I understand that you think this is a very small percentage," she told opponents of the plan, "but for these people, this is their life."

Confession: I hate public speaking.

The few readings I?ve done have been preceded by me telling my husband that fiction writers shouldn?t be expected to talk in front of other people, and that public speaking is a bizarre form of hazing. Nothing bad has happened?usually things go well?but they still make me nervous.

For the next one, I?m definitely going to reread Cynthia Leitich Smith?s post about how to handle public speaking events and potential problems like hecklers, not having enough time, and small turnout. Lots of really helpful advice and great anecdotes. I especially like her suggestion for how to politely respond to awkward or obnoxious questions.

Also, something very important to keep in mind about all kinds of author events:

With fall just around the corner ( yes it's wishful thinking!)? it will soon be time to start saving seeds from you garden. So I thought I would be real smart and make a seed template for you to print out. I?ve not made printables before and I searched high and low for a tutorial to learn how it's done. It took some work let me tell you. ?(to figure it out, for me at least) Here is the link to the one tutorial?that finally helped me in case you need one.

What I really want to do is learn how to create something as simple as a seed template on the computer. I had grandiose ideas of pretty vintage graphics decorating?mine but that was more than I could accomplish.??

I made this one (below) the old school way, drawing, tracing and cutting then scanning it into the computer. I scanned it as a photo then took it to Pixlr to add the text. It doesn?t look as slick as the other seed packet templates out there online. Is there anyone who can send me in the right direction to learn how to make one on my computer?
I made two printable seed packet templates for you to use if you like. One has a large seed packet on one page and the other has a small seed packet on one page for you to choose from. I tried to have two small packets on one page but for some reason Google would upload it as two pages with one packet on each. In Word I had two packets on one page. I have no idea what went wrong.

Here it is; they can be printed on regular printer paper or onto card stock for a sturdier packet. Pretty scrap book paper could also be used.

It would be a fun project for the kids to decorate your seed packet template before you cut it out. Let them color it with crayons, markers, or stamps if you like. I vote for small garden themed stamps J

Cut out your template and fold where you see the dotted lines in the picture. Your print out will not have the dotted lines. For a nice clean fold, use a ruler or other straight edge to fold the template over on. Glue back flaps and bottom flap down. Top can be taped or glued. If one day I learn to make a better and prettier seed packet template I'll replace this one for?you all.?And now that I have learned how to make a downloadable printable document, I can start giving you patterns on occasion?for the things I make. Like?for the?felt cupcakes I made a while back. I hope you enjoy my silly little seed packets.

  • If I feel the need for a change of scenery, I can take one of the interesting trips provided by the cruise company when we dock?at any of the exotic places they visit.
  • I was told that it takes months to get into a good retirement home but there is little delay in getting a reservation on a ship. What?s more, I can change to another of the company?s liners whenever I feel like it.
  • If my T.V. gives trouble or a light bulb needs replacing, they not only repair it fast, but apologize for the inconvenience.
  • There is a security guard on board, and every morning room service checks whether all the guests are doing well. If not, they send for a nurse or a doctor. If I am unfortunate enough to fall and break a hip, they will upgrade me to a suite for the rest of my life and? apologize profusely for any inconvenience I might have suffered.
  • Family will be only too keen to visit and might even check in for a mini-vacation. So, what more can a person ask for?

Federal Land Inc., the property unit of tycoon George Ty, has expressed interest in acquiring a foreclosed 8,000-square-meter prime lot in Makati City previously owned by a company of businessman Ramon Jacinto.

Federal Land executive vice president and general manager Jose Mari Banzon said in an interview the company had offered to acquire the property at the corner of Gil Puyat Ave. and Paseo de Roxas from Philippine National Bank.

Banzon said Federal Land planned to transform the lot into a mixed-use development.? The property is beside the head office of Ty-controlled Metropolitan Bank and Trust Co.

Banzon conceded that other property developers were also interested in acquiring the same lot, which served as an open parking lot since 1997.

Federal Land is a wholly-owned unit of GT Capital Holdings Inc., the listed holding company of the Ty group.

The property firm posted a net income of P1.7 billion in the first half, up 77 percent from previous year?s P201 million. Revenues grew 123 percent to P3.6 billion from P1.6 billion during the same period on higher real estate sales.

Federal Land has 19 ongoing projects and a landbank of 100.8 hectares as of -end 2011.

The Buendia property was previously subject to a dispute between PNB and RJ Group.

The storm-threatened Republican National Convention will cram four planned days of political theater into three by cutting some speakers, shortening some speeches, and starting evening speechifying earlier than previously planned, a senior strategist for Mitt Romney?s campaign told reporters on a conference call Sunday evening.

The convention will officially gavel in at 2 p.m. Monday but will be in session for no more than 10 minutes, said Russ Schriefer, the GOP strategist who has been the chief Romney campaign spokesman for convention logistics. The Republican National Committee called off Monday?s affairs due to the impending storm Isaac, which may be a hurricane by Monday as it blows past just west of Tampa, Fla., where tens of thousands of convention-goers and media representatives are gathering.

The proceedings will resume on Tuesday at 2 p.m. with Republicans taking their roll call vote in early evening to formally nominate Mr. Romney and his running mate, Rep. Paul Ryan (R) of Wisconsin, to the Republican ticket. The convention?s new schedule appears to have cut only one previously named speaker: Steve Cohen of Screen Machine, a portable crushing and screening equipment manufacturer. ?

However, convention planners offered a list of 15 GOP House members and House candidates plus three Senate candidates who could get less time at the podium in order to make way for those whom Mr. Schriefer dubbed ?headliners? originally scheduled to speak Monday. Among the headliners are Sen. Rand Paul (R) of Kentucky and former Arkansas Gov. Mike Huckabee, who will now speak Wednesday, along with South Carolina Gov. Nikki Haley and Texas GOP Senate candidate Ted Cruz, who will speak Tuesday.

The convention did not alter the most prime speaking space -- 10 p.m. to 11 p.m. -- on any of the remaining nights, Schriefer noted. Romney?s wife, Ann, and New Jersey Gov. Chris Christie (R) will headline Tuesday?s proceedings, followed by Representative Ryan on Wednesday and Sen. Marco Rubio (R) of Florida introducing Romney himself on Thursday. The evening speeches on Wednesday and Thursday will begin at 7 p.m. to fit in all speakers.

Schriefer also announced one other bit of convention showmanship: Republican National Committee Chairman Reince Priebus will start a clock counting how much debt the US accumulates every second when he gavels in the proceedings on Monday. When House Speaker John Boehner (R) of Ohio concludes the ceremonies on Thursday, he?ll stop the clock to demonstrate how much debt the nation racked up during the course of the GOP convention.?

Apple patent win may reshape sector, slow Google

Apple's decisive victory in a landmark US patent case against Samsung could reshape the hot sector for mobile devices and slow the momentum of Google and its Android system, analysts say.

Apple won more than $1 billion in the case Friday, after a California jury found the South Korean electronics giant infringed on dozens of patents held by the iPhone and iPad maker.

Although Google was not a party in the case, it makes the Android operating system which was central to the case -- a system which Apple's late co-founder Steve Jobs called a "stolen" product.

Apple has been battling as Samsung and other manufacturers of the free Android system eat away at its market share in the sizzling market for smartphones and tablet computers.

"I think this will force a reset on Android products as they are reengineered to get around Apple's patents," said Rob Enderle, a technology analyst and consultant with the Enderle Group.

But Enderle said other companies may benefit from the decision, including Microsoft, which has been lagging in the mobile sector, and Blackberry maker Research in Motion, which has been hit hardest by the rise of Android devices.

The court ruling, said Enderle, "should provide a stronger opportunity for both of Microsoft's new platforms -- Windows 8 and Windows Phone 8 -- because they come with indemnification against Apple, suddenly making them far safer and possibly a faster way to get product to market."

The decision also "will make RIM far more attractive as an acquisition because RIM's patents are thought to be strong enough to hold off Apple," Enderle said.

"Both Samsung and Google may make a play for the company, and both Microsoft and Apple may move to block them."

In recent months, Android devices have grabbed more than 50 percent of the US smartphone market to around 30 percent for Apple, while RIM's shares have slid to around 12 percent.

The patents at play include software such as the "bounceback" feature for smartphone users when scrolling and pinch-zooming, which are featured on Android devices.

Florian Mueller, a consultant who follows patent and copyright issues, said Friday's court ruling was "a huge breakthrough."

"The jury essentially concluded that Samsung is a reckless copycat and, since some of the infringement is Google's responsibility, basically agreed with Steve Jobs's claim that Android is a stolen product," he wrote on his blog.

Still, a lot hinges on what happens next in court, with the case likely to be tied up in appeal for some time.

Judge Lucy Koh set a September 20 hearing where she will consider whether to overturn or modify the jury verdict, whether to impose "punitive" damages which would triple the award and whether to issue injunctions against Samsung.

A critical factor will be whether Apple will be able to obtain a permanent injunction, or halt in sales on infringing Samsung devices, and whether this would be enforced during the appeal.

Dennis Crouch, a University of Missouri patent law specialist, said the judge will have broad discretion.

"Some courts have issued broad injunctions that essentially say 'stop infringing the patent,' others issue much more narrow orders directed only toward the particular products that are adjudged to infringe," Crouch said on his blog.

"The reality is that Samsung has been planning for the likelihood of injunctive relief and is surely ready to stop selling any of the infringing products and replace those products with ones that at least have not yet been adjudged as infringing."

This could lead to more legal battles, since Apple has another lawsuit pending on Samsung's newer handheld devices.

Samsung has pledged to keep fighting the case, and said that if it stands "it will lead to fewer choices, less innovation, and potentially higher prices."

Samsung called it "unfortunate that patent law can be manipulated to give one company a monopoly over rectangles with rounded corners, or technology that is being improved every day by Samsung and other companies."

Analysts say that aside from Samsung, Google could be the big loser, especially if Apple pursues its litigation against other manufacturers.

"Google cannot stop Apple. It is now on the run and will have to scramble to make software changes to Android," Mueller said.

"In a few years, the San Jose jury verdict may -- I repeat, MAY -- be remembered as the tipping point that sent Android on a downward spiral."

Romney assails Obama campaign on Akin, abortion - FOX19.com ...

TAMPA, Fla. (AP) - His Republican National Convention curtailed by a threatened hurricane, Mitt Romney conceded Sunday that fresh controversy over rape and abortion is harming his party and he accused Democrats of trying to exploit it for political gain.

"It really is sad, isn't it, with all the issues that America faces, for the Obama campaign to continue to stoop to such a low level," said Romney, struggling to sharpen the presidential election focus instead on a weak economy and 8.3% national unemployment.

His comments came as aides and party officials hurriedly rewrote the script for the convention, cut from four days to three because of the threat posed by approaching Tropical Storm Isaac. The storm is forecast to gain hurricane strength as it churns through the Gulf of Mexico but to pass well west of the convention city.

The revised schedule included a symbolic 10-minute session on Monday in a nearly empty hall, during which officials intend to launch a debt clock set to zero. The political objective is to show how much the government borrows throughout the convention week.

Officials did not rule out further changes because of the weather, and sidestepped when asked what might happen if, as seemed possible, the storm made landfall in the New Orleans area on the seventh anniversary of Hurricane Katrina. That storm killed 1,800 people and devastated the city.

"We're 100% full steam ahead on Tuesday," said Reince Priebus, chairman of the Republican National Committee, expressing confidence the 1-day delay would be the extent of the cancellations.

Despite concerns about the weather, a mammoth pre-convention celebration went on as planned Sunday night, attended by thousands of delegates and others who flocked to the Rays major league baseball stadium turned into a party venue in nearby St. Petersburg.

Priebus said Romney's nomination would take place on Tuesday, as would approval of a conservative party platform.

The former Massachusetts governor delivers his acceptance speech Thursday night before a prime time TV audience, then sets out on the final leg of a quest for the presidency that spans two campaigns and more than five years.

Polls make the race a close one, with a modest advantage for President Barack Obama.

For all the Republican attempts to make the election a referendum on the incumbent's handling of the economy, other events have intervened.

An incendiary comment more than a week ago by Rep. Todd Akin, the party's candidate for a Senate seat in Missouri, is among the intrusions. In an interview, he said a woman's body has a way of preventing pregnancy in the case of a "legitimate rape." The claim is unsupported by medical evidence, and the congressman quickly apologized.

Romney and other party officials, recognizing a political threat, unsuccessfully sought to persuade Akin to quit the race. Democrats have latched onto the controversy, noting not only what Akin said but also his opposition to abortion in all cases.

"Now, Akin's choice of words isn't the real issue here. The real issue is a Republican Party - led by Mitt Romney and Paul Ryan - whose policies on women and their health are dangerously wrong," said a recent letter from Debbie Wasserman Schultz, the chairwoman of the Democratic Party.

The party also posted a Web video that emphasizes the Republican Party's opposition to abortion and digitally alters the Republicans' "Romney-Ryan" logo to say "Romney Ryan Akin."

Interviewed on Fox, his comments broadcast on Sunday, Romney said the controversy over Akin "hurts our party and I think is damaging to women."

Romney spent the day in New Hampshire where he has a summer home. Aides said he was spending part of his afternoon practicing his convention speech with the use of a teleprompter.

Delegates marked time as the storm raked the Florida Keys to the south of the convention city en route to a projected landfall along the Gulf Coast.

"Somebody raised the prospect of marathon Monopoly. I favor the game Risk, but we'll see," said Tom Del Beccaro, chairman of the California delegation. I think people will just be ready for Tuesday and be pretty energetic then."

Hundreds of miles away, Romney said he was concerned for the safety of those who "are going to be affected" by the storm, which is predicted to worsen into a hurricane as it heads for landfall along the Gulf Coast.

In a presidential race defined by its closeness, Republican office-holders past and present said the party must find a way to appeal to women and Hispanics, and they said the economy was the way to do it.

"We have to point out that the unemployment rate among young women is now 16%, that the unemployment rate among Hispanics is very high, that jobs and the economy are more important, perhaps, than maybe other issues," said Arizona Sen. John McCain, who lost to Obama in 2008.

Former Florida Gov. Jeb Bush agreed, saying that Romney "can make inroads if he focuses on how do we create a climate of job creation and economic growth." If he succeeds, "I think people will move back towards the Republican side," Bush added.

Obama leads Romney among women voters and by an overwhelming margin among Hispanics, but he trails substantially among men.

The result is a race that is unpredictably close, to be settled in a small number of battleground states.

An estimated $500 million has been spent on television commercials so far by the two candidates, their parties and supporting outside groups, nearly all of it in Florida, North Carolina, Virginia, New Hampshire, Ohio, Iowa, Colorado and Nevada. Those states account for 100 electoral votes out of the 270 needed to win the White House.

Republicans have made no secret that they are eager to expand the electoral map to include Pennsylvania, Michigan, perhaps running mate Paul Ryan's Wisconsin and even Minnesota, states with 68 electoral votes combined.

All four are usually reliably Democratic in presidential campaigns. Yet Romney has a financial advantage over the president, according to the most recent fundraising reports, and a move by the Republicans into any of them could force Obama to dip into his own campaign treasury in regions he has considered relatively safe.

Making his case for the support of female voters, Romney said in the Fox interview: "'Look, I'm the guy that was able to get health care for all of the women and men in my state. ... 'I'm very proud of what we did."

It was a rare voluntary reference to the legislation he signed as governor of Massachusetts that required the state's residents to purchase coverage, the sort of mandate that is at the heart of Obama's federal legislation that conservatives oppose and Romney has vowed to see repealed.

Romney added that the state law was put into place "without cutting Medicare, which obviously affects a lot of women."

That was a reference to the federal law, which cut more than $700 billion in projected Medicare costs to help provide health care to millions who could not otherwise afford it.

Medicare generally favors Democrats as a political issue, but Romney has aggressively sought to cut into that advantage. He released a new television ad criticizing Obama's handling of the program with a catchphrase of "It ain't right."

The streets around the convention hall were crowded with police, National Guard and other security officials, who manned checkpoints in squads rather than individually.

A few hundred protesters gathered in a park about a half-mile from the convention vowed to make their point regardless of Tropical Storm Isaac. They set out large blocks of ice spelling out the words "middle class," and left them to melt on a warm, humid day, a gesture meant to signify middle class disappearance in a tough economy.

Bush and McCain were interviewed on NBC, and Priebus spoke on CNN.

Automate Your AS/400 With This Swiss Army Tool

When it comes to pulling out the Swiss army knife of all the tools available on the AS/400, iSeries and new IBM i midrange platforms you use a programming language called Control Language, or almost universally known as ?CL? to insiders.

Now when I say programming language, do not despair in believing you have to have some alphabet soup credentials or college programming wizard degree to be able to comprehend and utilize CL. It writes out as easily as a scripting language you may already be familiar with, and if you can string together phrases then you can write control language programs.

Do you enter commands directly onto the command line of the system today? Great, now you know how to write CL programs. Because any command you enter on a command line can be slapped together into a CL program with just a bar minimum of structuring and formatting.

One of my more favorite uses for CL is to fully automate tasks when used in conjunction with the included AS/400 job scheduler program. You don?t even need the fancy advanced job scheduler on the AS/400 or a third party program, the plain one will do.

Do you have a bunch of queries that are normally run by you each day? That is a perfect task to automate using CL and the run query command RUNQRY. Did you know that you can pass parameters to your Query/400 queries? Know with a little variable magic in your CL program you can pass a date parameter to your query.

Do you need several copies of the report to print in various places? CL has you covered by using the override with printer file command OVRPRTF you can create several copies of the spool file and move them to whatever output queues needed. You can even save the resultant report to a database file. How cool is that? CL makes it all happen.

One of the more nifty utilities I have put together using CL was a fully automated full system save backup routine. Using a simple little program it brought the AS/400 down to restricted mode by ending all of the subsystems, ran a save entire system exactly like using the ?go save 21? option onto the tape drive, executed a reclaim storage and more.

Now we are talking about some useful utilities that can be crafted. As you can see there is almost no limit to what can be done with CL.

To get started cranking out CL simply fire up the Program Development Manager with the STRPDM command and create a source physical file with the ?CLLE? type. Each CL program begins with STRPGM and ends with ENDPGM on a separate line, sandwiched in between those two lines of code you place your ordinary commands that you would otherwise enter on the command line. Pretty easy, huh?

When entering in your CL program if you get stuck on a command or forget a parameter for a specific command, not a problem. When editing a file using the source entry utility you can always press the F4 key to prompt a command and fill in the blanks, or in this case the parameters you want, and when you press enter the enter command with all the parameters you entered is validated and placed right into your source code file for you automagically.

Tropical Storm Isaac ? Don?t Forget About Your Pets

PRNews wire -?As Tropical Storm Isaac threatens to gather strength as it moves westward over the Caribbean Sea, The?Humane Society of the United States?reminds residents inPuerto Rico, the Caribbean islands,?Florida?and other Gulf states to prepare by taking some simple ? but critical ? steps to keep their pets safe.

While the path and impact of the storm is not certain, it could produce high winds, mudslides and flooding in low-lying areas. Residents in the potential areas of impact should stay up-to-date on the storm?s progress. Pet owners should have an emergency plan that includes the safety of their animals, and always be informed about the potential for evacuation in their area.

?It is crucial that residents are prepared to keep their pets inside if they are able to stay at home or to take pets with them if asked to evacuate in the face of this potentially destructive storm,? advised Niki Dawson, director of disaster services for The HSUS. ?If it isn?t safe for you, it isn?t safe for your pets.?

Dogs who are tethered as a means of confinement or other animals left outside may drown, choke to death on tangled leads, or suffer other serious injuries. Pets should be brought inside in the event of heavy downpours, storm surge or high winds. It?s also important to provide for your pets in the event you lose electricity, making certain they also have adequate food and water.

Things you can do right now:

  • Put a collar with visible identification on your pets, including indoor-only pets
  • Keep pictures of your pets on hand for identification purposes. Ideally, you should also be in the photo
  • Create a pet emergency kit (see below) and refresh the items every few months
  • Talk to your neighbors about how they can help your pets if you are not at home when disaster strikes
  • Create a list of hotels that allow pets
  • Plan on evacuating about 100 miles inland

This emergency supply kit should include:

  • Three-or-more-day supply of food in airtight, waterproof container, and drinking water
  • Bowls for food and water
  • Current photos and physical description of your pets, including details on markings
  • Medications, vaccination records and pet first aid supplies
